This study examined the dose response of perindopril in hypertension, and inclusion criteria required participants to have supine diastolic blood pressure between 95 and 114 mm Hg after a 4-week washout period.
Results of a randomized, double-blind, placebo-controlled, parallel-group study of 293 patients in which 57 patients were assigned to the 4 mg group and 59 patients to the 8 mg group. Of these, 55 were assessed for efficacy at 12 weeks of active treatment in the 4 mg group and 54 in the 8 mg group (P<0.05 vs baseline and placebo). 3
Placebo-corrected changes in BP measured 24 hours postdose (trough effect) for the 4 mg and 8 mg doses were: systolic -5 mm Hg and -11 mm Hg, respectively, and diastolic -6 mm Hg and -8 mm Hg, respectively 3

In controlled trials, ACE inhibitors have an effect on blood pressure that is less in black patients than in nonblacks. Also, black patients receiving ACE inhibitor monotherapy have been reported to have a higher incidence of angioedema than do nonblacks. 2
Like other ACE inhibitors, ACEON® can cause symptomatic hypotension. ACEON® has been associated with hypotension in 0.3% of uncomplicated hypertensive patients in US placebo-controlled trials. Symptoms related to orthostatic hypotension were reported in another 0.8% of patients. This is most likely to occur in patients who have been volume and/or salt-depleted. This should be corrected before initiating therapy with ACEON® (see DOSAGE AND ADMINISTRATION).2
